Factors That Affect Streptomyces Growth Biology Essay The growth of a microorganism is extremely dependent upon the surrounding environmental conditions. Every organism has a range of optimum conditions at which members of its species will grow ideally. Inoculum, temperature, pH and dissolved oxygen concentration (among other factors) within the media is often carefully controlled in industrial applications as deviations from the optimal conditions often have profound effects on the growth rate and activity of the organism (Shuler and Kargi, 1992). When directing experiments involving microbial growth, the preparation and quality of the inocula are as important as any other parameter control. In Streptomyces if mycelia are used in the inoculum, many uncontrolled variables will emerge. These variables include age, stress level and growth stage of the cells, all of which obligatory should be controlled (Kieser et al., 2000). This problem gets even worse when attempting to initiate trials over a period of several months as even small variations in the population size of the inoculum can result in large changes in the level and rate of growth. Streptomyces vegetative inocula are often prepared through the addition of several colonies sometimes from many starter plates to small amounts of media. This is afterward incubated until sufficient biomass is produced, as determined through optical density measurements of the media. This method of vegetative inoculum used by Jakeman et al. (2004) is effective in obtaining biomass; neverthele ss it does not provide the consistent inoculum required for comparative trials. The number of cells within the inoculum is hard to directly measure and the inconsistent manner of initially adding cells can result in cells at different stages of growth or stress level. Another method of preparing an inoculum is to harvest spores from the selected culture and store under conditions not satisfactory for germination. The cells within a spore suspension remain in a consistent state while still providing an instant inoculum (Kutzner and Nitsch, 1970). This technique also allows long term storage of a viable inoculum that should produce reliable results. By suspending the spores in a soft agar spore suspension, Kutzner and Nitsch (1970) have proven that a suspension of Streptomyces spores will remain viable as an inoculum over a year. Temperature The bacterial growth behavior is greatly influenced by many factors such as environmental conditions and the physiological state of the bacteria. The temperature of the environment directly affects the activity and growth of cells; every species has an ideal temperature for growth that is influenced by its physiology (Shuler and Kargi, 1992). This optimum temperature for growth may not be the same as the temperature at which metabolites are produced most efficiently. Streptomyces venezuelae ISP 5230 are mesophilic bacteria, which means that their optimal growth temperature is between 15 and 40 Â °C. For every increase of 10 Â °C closer to the optimum growth temperature, the growth rate of bacteria approximately doubles (Truelstrup-Hansen et al, 2002). If the media temperature is above optimal, the rate of growth is reduced. The temperatures used for Streptomyces venezuelae growth have been cited as 30 Â °C Â ± 2 Â °C (Wang and Vining, 2003; Jakeman et al., 2006). At 42 Â °C Str eptomyces venezuelae express heat shock genes that motivate sporulation and antibiotic production (Yang et al., 2008). pH Similarly to temperature, the pH (hydronium ion concentration) of the media affects the growth rate of bacteria by influencing the activity of cellular enzymes (Shuler and Kargi, 1992). To achieve the maximum growth rate level, current literature recommends growing Streptomyces venezuelae in a media with the maintenance of pH between 6.0 7.8 (Jakeman et al., 2006). The optimal pH levels are often different when maximizing growth rate and highest product yield (Shuler and Kargi, 1992). However, bacteria are often able to grow within a huge range of pH. Studies on the Streptomyces species by Kontro et al. (2005) have found the optimal pH ranges of growth and sporulation for this species to be between 5.5 and 11.5. The study executed by Kontro et al. (2005) has also found that the media strongly affects the acid tolerance of Streptomyces species. When grown at pH 4 in a media that contains yeast extract most species included in the studies were unable to grow, while at the pH of 7 the majority of species displayed maximum growth rate on most types of media. In order to buffer the pH of the growth medium to near neutral values, a buffer called MOPS (3-(N-morpholino) propanesulfonic acid) is normally used for this species of Streptomyces. While studying the effects of carbon sources on antibiotic production by Streptomyces venezuelae, MOPS was added to jadomycin B production media by Jakeman et al. (2006) however, no buffer was added to the Maltose yeast extract malt extract (MYM) agar that was used for the growth during the study. Glazebrook et al. (1990) studied the effect of different carbon sources on Streptomyces venezuelae sporulation in minimal LS media. In this media, enough amounts of MOPS were added to maintain the pH of the media at near neutral values for every carbon source except glucose. During growth in glucose-based LS media, a high production of oxo-acids was observed resulting in growth inhibition. Dissolved Oxygen The dissolved oxygen available to bacterial cells within the media can be a limiting factor if the rate of consumption exceeds the rate of supply (Schuler, 2000). There is a critical oxygen concentration exclusive to each bacteria species, above which the growth rate is independent of the media dissolved oxygen concentration. This value is usually 5 10 % of the maximum saturation level; however the maximum saturation is influenced through the existence of dissolved salts, organics, and the medias temperature (Shuler and Kargi, 1992). The production of secondary metabolites can also be influenced through dissolved oxygen concentration. During antibiotic production Streptomyces clavuligerus, increased oxygen saturation above the critical dissolved oxygen concentration which resulted in additional product and biomass yield (Yegneswaran et al., 1991) up to the maximum saturation limit of oxygen in the media. Substrate The substrate used in the media is an important factor that can affect growth rate of the bacteria as well as its characteristics. For instance, Kontro et al. (2005) found that Streptomyces species changed its behavior as acidophiles, neutrophiles or alkalophiles, depending on the growth media. Most species would grow optimally at pH that is near to neutral or slightly alkaline conditions on a media that included yeast extract (Kontro et al., 2005). Streptomyces venezuelae is grown using many carbon sources including glycerol, glucose, lactose, mannose, maltotriose, maltose starch and more. The effect of these carbon sources on growth and secondary metabolite production is dependent upon many factors such as pH (Kontro et al., 2005). For example, studies on the sporulation of Streptomyces venezuelae in liquid media by Glazebrook et al., (1990) found that while maltose supported the growth and sporulation of Streptomyces venezuelae, glucose significantly inhibited sporulation. Specifi cally, when grown in minimal LS media using glucose as the main carbon source, the pH of the media dropped rapidly with the production of oxo-acids by the bacteria. The increased acidity likely contributed to growth inhibition; however the glucose was reported to also have an inhibitory effect on sporulation that was un-related to the increased acidity of the media. As the jadomycin family of antibiotics is linked to the sporulation pathway (Wang and Vining, 2003), it is visible that the media has a huge effect on the growth and antibiotic production by Streptomyces venezuelae. Work by Jakeman et al. (2006) found that the carbon source supplied in to the media also has an influence on the antibiotic production by Streptomyces venezuelae. The effect of several sugars on jadomycin B production was studied and important differences in antibiotic production between sugars were found, where glucose was found to be optimal. Heat Balance The bacterial behavior is greatly influenced by the environmental temperature; however the metabolism and activity of bacteria also affects media temperature. Naturally about 50 60 % of energy produced during the metabolism of nutrients within a system is transformed into heat energy (Shuler and Kargi, 1992). The metabolism of sugar is a great source of thermal energy that must be taken into consideration in bioreactor design (Ben-Hassan et al., 1992). Streptomyces venezuelae aerobically metabolize sugars for energy and the production of metabolites. In industry, the amount of substrate used by the microorganism for growth and product synthesis determines the economic viability of the process. The heat energy produced can be determined through the utilization of a heat balance model (Ghaly et al., 1992). Once the heat of mixing is determined, the energy generated by Streptomyces venezuelae can be determined. The work by Ghaly et al. (1992) on the heat generated through mixing and me tabolism during lactose fermentation showed that the temperature of media increased from 22 to 32 Â °C with bacterial growth (Figure 3.9). This increase in temperature was due to the heat generated by the yeast (7.4 JÂ ·s-1) as well as the heat of mixing (1.01 JÂ ·s-1). The amount of heat generated by the organisms in a bioreactor can be estimated by modeling the heat flux over the boundary of the enclosed system. This mathematical model or heat balance balances the thermal energy generated within a system with the energy stored and lost from the system (Ben-Hassan et al., 1992). To develop a heat balance using this method, a trial must first be executed on the system through normal stable state operation to evaluate the heat generation during operation of the bioreactor. A heat balance can be performed on a bioreactor to determine the amount of thermal energy generated within the system during normal operation due to mixing. To model the system under different operating circumst ances the reactor can be run at several agitation speeds (e.g. 200, 400 and 600 rpm) until a state of thermal balance is reached. At thermal balance the amount of energy generated within the bioreactor is equal to the amount of energy lost; this state of stability can be used to eliminate unknown parameters within the heat balance model.

Moral Theories A.  Ã‚  Ã‚  Ã‚  Ã‚  Morality comes from God. Therefore, moral behavior is that behavior that conforms to the will of God. Immoral behavior defies the will of God. The will of God is correctly interpreted by the Church. Rating: 6. I was feeling this one, until the last sentence. There are so many denominations, and the reason is that they disagree with one another. Many religions interpret what the will of God is differently, not to mention incorrectly. I do believe that a big part of morality comes from God. B.  Ã‚  Ã‚  Ã‚  Ã‚  Do unto others as you would have them do unto you. Rating:1! Ahhh, the Golden Rule-you gotta love it. I suppose the only problem is like the example in the book of the African tribe that eats their dead, if I died there, they would eat me because they would want to be eaten. Very tricky. C.  Ã‚  Ã‚  Ã‚  Ã‚  Be whatever you are; do whatever you want to do—just as long as you don’t hurt anybody. Rating:7. This sounds to me like humanism, although I’m not sure anymore if I know what that is. Moral values come from the laws and norms of the society. Therefore, what is morally right in one society could be morally wrong in another. Rating 4. Doesn’t the answer to this one really decide if you are an objectivist or realist? My husband and I argued this one, and he is a strong objectivist. I am caught in the middle because I don’t understand how you can truly impose your beliefs on people that have no idea what we consider to be right. What if people tried to do that to us? F.  Ã‚  Ã‚  Ã‚  Ã‚  People never act immorally deliberately. When a person acts immorally, it is because he or she has a mistaken notion of what is truly good. Immoral behavior is ignorance or stupidity, not wickedness. Rating 9. Whoa, do I disagree with this one. I do believe many people do wrong things because of ignorance, but there are many people who do things wrong and they do it knowing that it is wrong.

Child Devlopment Essay

Cognitive Development consists of imagination, sequencing, problem solving and memory, all these are featured in the boy I observed, Aaron King who was the little boy I chose to observe. He is a four year-old preschooler at the above named learning center. I started observing Aaron at the beginning of their circle time. During the circle time, Aaron amongst other fourteen kids in his classroom, sang the popular American Song: â€Å"IF YOU ARE HAPPY AND YOU KNOW IT, CLAP YOUR HANDS†¦. † While singing this song, the children’s voices were up and loud which indicated that they enjoyed singing songs. Aaron’s participation was great, he was so excited about the choruses, clapping & stomping of his feet. All the children in turn repeated and imitated all the actions of the teacher; as she needed the audience and focus of the kids for easy understanding of the wordings including choruses of the song. Aaron standing close to two other kids, a boy and a girl sang the song with seriousness while clapping their hands & stomping their feet at the same time. The teacher taught the Safety signs briefly. The children’s literacy is very high while learning safety signs as they were very interested in the emphasis laid in the safety signs. In order to ensure that the children understood what they were taught, the teacher asked Aaron what a particular sign stood for which he answered correctly. For example, stop sign is signified by a red color while â€Å"no bicycle† sign is signified by a cross on a bicycle, so on and so forth. The teacher taught the kids about â€Å"Me† book which described the parts of the body, however, she asked Aaron to touch his hands, elbows & cheek which he did correctly. After singing the clapping song and some other songs, the teacher changed the activity to coloring books. Meanwhile, Aaron moved from one place to another. He jumped and hopped around the classroom showing excitement. At a time, the teacher had to sit him down â€Å"Aaron sit down and do your coloring†¦ † He sat down to color while another boy watched him doing the coloring. Bryan & Joy who sat together sharing the same table with Aaron were also coloring; they also talked to one another in friendly manner while looking at each other’s work. Aaron was as excited as he was coloring his book. He finished his coloring using nice colors to color different animals indicating that he had a good taste. He also made a ship with legos, I asked him why he made a ship, and he told me that he would like to be an architect. Aaron amongst others was able to write their names and figure out pictures on the paper. For example, the letter Bb for ball and drew a line to join the letter to the picture of ball. He was able to do that for all the alphabetic letters to compliment individual pictures on the paper. Soon, Aaron stood up to go to the library; as he got to the place where the inscription of â€Å"Quiet Zone† was, another boy ran towards him and pushed him together with the book that he just picked – â€Å"Alphabet adventure†. He staggered, but he immediately got control of himself while he looked so surprised that he was pushed without doing anything. He reported the matter to the teacher â€Å"Ms. Patty, Josiah push(ed) me†¦ † then the teacher told Josiah to say sorry to Aaron which he did, then they both hugged one another. Thereafter, he went to sit down quietly in the library corner to read his book. He opened up the book so fast as if he knew where to read, then he stirred at a page that really caught his attention and started reading quietly. In summary, Cognitive Development is a very important outcome of learning for preschoolers as they always have their brain developed through creative arts. This consists of imagination, sequencing, problem solving and memory; all these are featured in the boy I observed. Acting drama is acting another character by putting on the costumes – for example, acting as a professional doctor or a lawyer or a nurse can be an eye opener to the preschoolers on what profession they may aspire to be in the near future; Aaron wants to be an architect, he says he likes to put up beautiful buildings. A good imaginative skill is very important for preschoolers to become smart; similarly, sequencing – doing things in an orderly manner is also a good learning skill. Ultimately, Aaron Kingsland has all these qualities. Observer’s Name: —————————————- Location:Early Creativity Learning Center Classroom LANGUAGE & LITERACY DEVELOPMENT OBSERVATION OF A PRESCHOOLER – 4 YEARS OLD. My observation of language and literacy in a preschool classroom for children of four and five years old was interesting. Aaron Kingsland (four years old) was the preschooler at the above named learning center whom I chose to observe. I started observing Aaron at the beginning of their circle time. The kids in the classroom were seventeen in number – eight girls and nine boys. I observed Aaron in particular in the class for two and half hours from the beginning of the circle time till the end. The primary language used by the teacher was English; Aaron also speaks same language as well as the whole class. Language is said to be a means of communicating ideas, & feelings. There are four basic developmentally appropriate practices and clear language outcomes in the preschool classroom I observed. They are Literacy Development, Intellectual, Physical, Social and Emotional Developments. Literacy is a very important developmental appropriate practice. The four basic skills which young children need are Speaking, Listening, Writing and Reading. These were ultimately displayed by Aaron in the circle time activities performed that morning. Reading, talking and expressing what they do and following directions go a long way for preschoolers Through reading, Aaron amongst other peers has book knowledge, comprehension and appreciation of what he read. More importantly, literacy goes a long way in our society and even in the world.

Part Three Chapter X

X Parminder worked late on Monday evenings, and as Vikram was usually at the hospital, the three Jawanda children laid the table and cooked for themselves. Sometimes they squabbled; occasionally they had a laugh; but today, each was absorbed in their own particular thoughts, and the job was completed with unusual efficiency in near silence. Sukhvinder had not told her brother or her sister that she had tried to truant, or about Krystal Weedon's threat to beat her up. The habit of secrecy was very strong in her these days. She was actively frightened of imparting confidences, because she feared that they might betray the world of oddness that lived inside her, the world that Fats Wall seemed able to penetrate with such terrifying ease. All the same, she knew that the events of the day could not be kept quiet indefinitely. Tessa had told her that she intended to telephone Parminder. ‘I'm going to have to call your mum, Sukhvinder, it's what we always do, but I'm going to explain to her why you did it.' Sukhvinder had felt almost warm towards Tessa, even though she was Fats Wall's mother. Frightened though she was of her mother's reaction, a tiny little glow of hope had kindled inside her at the thought of Tessa interceding for her. Would the realization of Sukhvinder's desperation lead, at last, to some crack in her mother's implacable disapproval, her disappointment, her endless stone-faced criticism? When the front door opened at last, she heard her mother speaking Punjabi. ‘Oh, not the bloody farm again,' groaned Jaswant, who had cocked an ear to the door. The Jawandas owned a patch of ancestral land in the Punjab, which Parminder, the oldest, had inherited from their father in the absence of sons. The farm occupied a place in the family consciousness that Jaswant and Sukhvinder had sometimes discussed. To their slightly amused astonishment, a few of their older relatives seemed to live in the expectation that the whole family would move back there one day. Parminder's father had sent money back to the farm all his life. It was tenanted and worked by second cousins, who seemed surly and embittered. The farm caused regular arguments among her mother's family. ‘Nani's gone off on one again,' interpreted Jaswant, as Parminder's muffled voice penetrated the door. Parminder had taught her first-born some Punjabi, and Jaz had picked up a lot more from their cousins. Sukhvinder's dyslexia had been too severe to enable her to learn two languages and the attempt had been abandoned. ‘†¦ Harpreet still wants to sell off that bit for the road †¦' Sukhvinder heard Parminder kicking off her shoes. She wished that her mother had not been bothered about the farm tonight of all nights; it never put her into a good mood; and when Parminder pushed open the kitchen door and she saw her mother's tight mask-like face, her courage failed her completely. Parminder acknowledged Jaswant and Rajpal with a slight wave of her hand, but she pointed at Sukhvinder and then towards a kitchen chair, indicating that she was to sit down and wait for the call to end. Jaswant and Rajpal drifted back upstairs. Sukhvinder waited beneath the wall of photographs, in which her relative inadequacy was displayed for the world to see, pinned to her chair by her mother's silent command. On and on went the call, until at long last Parminder said goodbye and cut the connection. When she turned to look at her daughter Sukhvinder knew, instantly, before a word was spoken, that she had been wrong to hope. ‘So,' said Parminder. ‘I had a call from Tessa while I was at work. I expect you know what it was about.' Sukhvinder nodded. Her mouth seemed to be full of cotton wool. Parminder's rage crashed over her like a tidal wave, dragging Sukhvinder with it, so that she was unable to find her feet or right herself. ‘Why? Why? Is this copying the London girl, again – are you trying to impress her? Jaz and Raj never behave like this, never – why do you? What's wrong with you? Are you proud of being lazy and sloppy? Do you think it's cool to act like a delinquent? How do you think I felt when Tessa told me? Called at work – I've never been so ashamed – I'm disgusted by you, do you hear me? Do we not give you enough? Do we not help you enough? What is wrong with you, Sukhvinder?' In desperation, Sukhvinder tried to break through her mother's tirade, and mentioned the name Krystal Weedon – ‘Krystal Weedon!' shouted Parminder. ‘That stupid girl! Why are you paying attention to anything she says? Did you tell her I tried to keep her damn great-grandmother alive? Did you tell her that?' ‘I – no – ‘ ‘If you're going to care about what the likes of Krystal Weedon says, there's no hope for you! Perhaps that's your natural level, is it, Sukhvinder? You want to play truant and work in a cafe and waste all your opportunities for education, because that's easier? Is that what being in a team with Krystal Weedon taught you – to sink to her level?' Sukhvinder thought of Krystal and her gang, raring to go on the opposite kerb, waiting for a break in the cars. What would it take to make her mother understand? An hour ago she had had the tiniest fantasy that she might confide in her mother, at last, about Fats Wall †¦ ‘Get out of my sight! Go! I'll speak to your father when he comes in – go!' Sukhvinder walked upstairs. Jaswant called from her bedroom: ‘What was all that shouting about?' Sukhvinder did not answer. She proceeded to her own room, where she closed the door and sat down on the edge of her bed. What's wrong with you, Sukhvinder? You disgust me. Are you proud of being lazy and sloppy? What had she expected? Warm encircling arms and comfort? When had she ever been hugged and held by Parminder? There was more comfort to be had from the razor blade hidden in her stuffed rabbit; but the desire, mounting to a need, to cut and bleed, could not be satisfied by daylight, with the family awake and her father on his way. The dark lake of desperation and pain that lived in Sukhvinder and yearned for release was in flames, as if it had been fuel all along. Let her see how it feels. She got up, crossed her bedroom in a few strides, and dropping into the chair by her desk, pounded at the keyboard of her computer. Sukhvinder had been just as interested as Andrew Price when that stupid supply teacher had tried to impress them with his cool in computing. Unlike Andrew and a couple of the other boys, Sukhvinder had not plied the teacher with questions about the hacking; she had merely gone home quietly and looked it all up online. Nearly every modern website was proof against a classic SQL injection, but when Sukhvinder had heard her mother discussing the anonymous attack on the Pagford Parish Council website, it had occurred to Sukhvinder that the security on that feeble old site was probably minimal. Sukhvinder always found it much easier to type than to write, and computer code easier to read than long strings of words. It did not take very long for her to retrieve a site that gave explicit instructions for the simplest form of SQL injection. Then she brought up the Parish Council website. It took her five minutes to hack the site, and then only because she had transcribed the code wrong the first time. To her astonishment, she discovered that whoever was administering the site had not removed the user details of The_Ghost_of_Barry_Fairbrother from the database, but merely deleted the post. It would be child's play, therefore, to post in the same name. It took Sukhvinder much longer to compose the message than it had to hack into the site. She had carried the secret accusation with her for months, ever since New Year's Eve, when she had noticed with wonder her mother's face, at ten to midnight, from the corner of the party where she was hiding. She typed slowly. Autocorrect helped with her spelling. She was not afraid that Parminder would check her computer history; her mother knew so little about her, and about what went on in this bedroom, that she would never suspect her lazy, stupid, sloppy daughter. Sukhvinder pressed the mouse like a trigger.
